Filmul începe la aproximativ un an de la evenimentele din Spider-Man: Into the Spider-Verse, cu Miles Morales (Shameik Moore) bucurându-se de rolul său de Spider-Man, dar luptându-se să-și echilibreze viața normală și îndatoririle sale de super-erou. Când Gwen Stacy (Hailee Steinfeld) se întoarce în universul lui Miles, el descoperă că multiversul a fost cu adevărat larg deschis în urma bătăliei sale cu Kingpin, iar acum o nouă amenințare, The Spot (Jason Schwartzman), amenință întregul multivers.
